Aspergillus oryzae extracts are rich in prolyl endopeptidases and dipeptidyl peptidase IV (DPP-IV). DPP-IV is a multifunctional serine protease that targets proline-containing peptides, such as those found in gliadin and casein, two proteins resistant to proteolysis. The protease activity of Aspergillus oryzae targets the proline-rich 33-mer of gliadin, perhaps the most immunotoxic peptide derived from gliadin. The antigenic and immunomodulatory proline-rich proteins in gluten are thought to trigger an inflammatory response in individuals with celiac disease or gluten sensitivity. This reaction is often due to inadvertent exposure to gluten, even in individuals following a gluten-free diet. Aspergillus oryzae protease supplementation has also been associated with increased abundance of Bifidobacterium and Lactobacillus in experimental models, suggesting a potential probiotic effect.
